Playing as Luigi in single player: Hold down the L and R shoulder buttons and select one of your saved game files. You'll then be able to start playing as Luigi.
Playing in secret Challenge mode: Select one of your saved game files. Then pause the game on the world map and press LRLRXXYY. Then you can play in secret Challenge mode.
